Use of rotavirus virus-like particles as surrogates to evaluate virus persistence in shellfish ArchiMer

Rotavirus virus-like particles (VLPs) and MS2 bacteriophages were bioaccumulated in bivalve mollusks to evaluate viral persistence in shellfish during depuration and relaying under natural conditions. Using this nonpathogenic surrogate virus, we were able to demonstrate that about 1 log(10) of VLPs was depurated after 1 week in warm seawater (22 degrees C). Phage MS2 was depurated more rapidly (about 2 log(10) in 1 week) than were VLPs, as determined using a single-compartment model and linear regression analysis. After being relayed in the estuary under the influence of the tides, VLPs were detected in oysters for up to 82 days following seeding with high levels of VLPs (concentration range between 10(10) and 10(9) particles per g of pancreatic tissue)...

Potential of F-specific RNA bacteriophages to discriminate sources of faecal pollution in French shellfish ArchiMer

In Europe, shellfish harvesting areas are classified according European Directive EC N°854/2004, using classic faecal indicator levels (Escherichia coli). A better indicator is needed, however, because shellfish may contain other human pathogens. Other indicator organisms have therefore been evaluated and, among these, coliphages meet most of the requirements to serve as indicators and models for enteroviruses. Identification of 129 Micrococcaceae strains isolated from food of animal origin ArchiMer

129 strains of Micrococcaceae were isolated from food of animal origin (minced meat, cakes with confectioner's custard) on Baird-Parker medium. 120 were Staphylococcus and 9 Micrococcus, according to the ID 32 STAPH-System (1989). The results were analyzed using a computer program. 60% of these 120 Staphylococcus strains of animal origin were coagulase positive S. aureus (aurease+, Rapidec Staph technique). 46% were resistant to novobiocin (5 microg/ml), 13 to 44% were identified with negative discordant tests using this micromethod. 44% produced no acetoin, 35% had no urease and 15% no arginine dihydrolase. 48 Staphylococcus strains (40%) were identified as human coagulase negative Staphylococcus species (strains: 39; species: 9) or as animal species...

An inter-laboratory study of the accuracy of microbial source tracking (MST) methods was conducted using challenge fecal and sewage samples that were spiked into artificial freshwater and provided as unknowns (blind test samples) to the laboratories. The results of the Source Identification Protocol Project (SIPP) are presented in a series of papers that cover 41 MST methods. Evaluation of two library-independent microbial source tracking methods to identify sources of fecal contamination in french estuaries ArchiMer

In order to identify the origin of the fecal contamination observed in French estuaries, two library-independent microbial source tracking (MST) methods were selected: (i) Bacteroidales host-specific 16S rRNA gene markers and (ii) F-specific RNA bacteriophage genotyping. The specificity of the Bacteroidales markers was evaluated on human and animal (bovine, pig, sheep, and bird) feces. Two human-specific markers (HF183 and HF134), one ruminant-specific marker (CF193'), and one pig-specific marker (PF163) showed a high level of specificity (>90%). Les études font généralement état d'une lente dégradation de l'environnement marin avec diminution des coraux et de la faune associée, au profit d'une augmentation de la couverture algale. Seismic, Seabeam, magnetic and gravimetrie surveys in the southern part of the N-Fiji basin were conducted during the SEAPSO LEG III Cruise. The active spreading axis was recognized between 17°30'S and 21°S. It consists of a continuous N-S Ridge cross-cut by transverse directions oriented N25 and N45. North of 17°30'S, the spreading System is more complex due to the existence of a possible triple junction located around 15° S. The eastern part of the N-Fiji basin, west of Viti Levu is mostly characterized by alternating highs and deeps (<4,000 m). This area must be regarded no more as a spreading center, but as complex boundary suffering transverse tectonics.
